چکیده

A class of acidic nuclear phosphoproteins has been isolated throughout the mitotic cycle and at two points during differentiation in the slime mold Physarum polycephalum. The electrophoretic profiles of these proteins are reproducible and unchanging throughout the mitotic cycle, but reproducible changes occur during differentiation. The proteins are rapidly synthesized after mitosis, and their molecular weights range from 34,000 to 88,000. The proteins rapidly incorporate [(32)P]orthophosphate, and the content of alkali-labile phosphate increases 20 percent during the period after DNA synthesis. The proteins comprise 6.5 percent by dry weight of nuclear material while the DNA comprises about 5.5 percent. These acidic nuclear proteins may have a role in control of gene activity.

Change in properties of Physarum polycephalum amoebae during extended culture.

During two years of passage, several myxamoebal strains of Physarum polycephalum changed with regard to their properties in crosses and plaque morphology. These changes have been correlated with increased nuclear deoxyribonucleic acid content.

DNA methylation during differentiation of a lower eukaryote, Physarum polycephalum.

Starvation-induced differentiation of the slime mould Physarum polycephalum is accompanied by continuous methylation of DNA. No stable changes in the overall level of DNA methylation are evident, but a gene known to be transcribed specifically during differentiation is subject to increased methylation. Inhibitors of DNA methylation preclude differentiation of P. polycephalum, although they are ...

Synthesis and some properties of an actin-like nuclear protein in the slime mold Physarum polycephalum.

A protein was extracted from isolated nuclei of the slime mold Physarum polycephalum which could be labeled with radioactive precursors only during G(2) phase. The native protein was purified by extraction in low-ionic-strength buffer [10 mm tris(hydroxymethyl)aminomethane-hydrochloride] of isolated nuclei and by preparative polyacrylamide gel electrophoresis. It was extracted from isolated nuc...

A Next-Generation Sequencing Approach to Study the Transcriptomic Changes During the Differentiation of Physarum at the Single-Cell Level

Physarum polycephalum is a unicellular eukaryote belonging to the amoebozoa group of organisms. The complex life cycle involves various cell types that differ in morphology, function, and biochemical composition. Sporulation, one step in the life cycle, is a stimulus-controlled differentiation response of macroscopic plasmodial cells that develop into fruiting bodies. Well-established Mendelian...

Biosynthesis and posttranslational acetylation of histones during spherulation of Physarum polycephalum.

Plasmodia of Physarum polycephalum can be induced to differentiate into dormant spherules: DNA-, RNA- and protein-synthesis cease during this process. Analysis of the histone H4 acetylation during spherulation revealed no significant changes of the relative acetate content and percentage of acetylated H4 subspecies. This result does not support a close correlation of histone acetylation and tra...
